Vascular diseases are major causes of morbidity and mortality. Vascular smooth muscle cells (VSMCs), as integral constituents of the vascular wall, provide structural support and dynamically alter vessel shape and structure in response to environmental stimuli. While the role of their endothelial neighbors has been extensively examined in facilitation of ischemic responses and vascular remodeling, less is known about VSMCs. Hypoxia-inducible factors (HIFs) are master regulators of cellular responses to O2 tension. These heterodimeric transcription factors drive endothelial vascularization and are putative regulators of VSMC driven vascular remodeling in pulmonary artery hypertension. The studies presented in this dissertation examine the role of canonical HIF activity in modulating VSMC phenotype, vasoreactivity, and responses to peripheral ischemia. Mice with smooth muscle specific deletion of aryl hydrocarbon receptor nuclear translocator (ARNT; HIF-1ß) (ArntSMKO), required for HIF transcriptional activity, feature impaired peripheral perfusion recovery following femoral artery ligation. Vessels from ArntSMKO mice display altered vasomotor function, increased hypoxic microvascular permeability, and disrupted arteriolar VSMC morphology. Aberrant behavior and transcriptional profile in isolated ArntSMKO VSMCs indicate phenotypic switching. Collectively, the data support a role for ARNT regulating VSMC phenotype, essential for vasoreactivity and perfusion recovery in peripheral ischemia. As such, ARNT provides a therapeutic target for modulating VSMC phenotypic change, a feature of many vascular diseases.

"Over the last three decades, lionfish populations have exploded throughout the western Atlantic, Caribbean Sea, and Gulf of Mexico. Their high abundances and gluttonous appetites threaten not only ecosystems like coral reefs, but also important nursery areas, and deep habitats hundreds of feet below diving depths. In some shallow water dive sites, spearfishing has proven an effective way to control populations. For those locations, sustained removals may limit the impacts these fish on the invaded ecosystems. Numerous novel devices and approaches to harvest lionfish from water depths beyond recreational scuba limits have been considered, but none have yet proven successful. Developing an effective method to target lionfish from these and other remote locations is important. Recent studies have reported the negative effects of lionfish on important prey species of both recreationally and commercially important species in deep water. There may also be trap applications in some shallow water locations, like nurseries, where lionfish control would protect the juveniles of some valuable fished species, and other ecologically important species"--Page 1.

This chapter reviews recent (2000-2005) personality disorder (PD) research, focusing on three major domains: assessment, comorbidity, and stability. (a) Substantial evidence has accrued favoring dimensional over categorical conceptualization of PD, and the five-factor model of personality is prominent as an integrating framework. Future directions include assessing dysfunction separately from traits and learning to utilize collateral information. (b) To address the pervasiveness and extent of comorbidity, researchers have begun to move beyond studying overlapping pairs or small sets of disorders and are developing broader, more integrated common-factor models that cross the Axis I-Axis II boundary. (c) Studies of PD stability have converged on the finding that PD features include both more acute, dysfunctional behaviors that resolve in relatively short periods, and maladaptive temperamental traits that are relatively more stable, similar to normal-range personality traits, with increasing stability until after 50 years of age. A new model for assessing PD and perhaps all psychopathology emerges from integrating these interrelated reconceptualizations.
