The transmembrane photosynthetic protein complex Photosystem I (PSI) possesses remarkable photoactive electrochemical properties which make it highly sought after for incorporation into biohybrid photovoltaic devices. In the pursuit of all such endeavors, three main factors must always be kept in view: understanding the direct redox transfer steps, threedimensional coordination and stabilization of PSI aggregates, and interfacial connectivity with conductive pathways. This thesis presents steps taken to further these three concepts, contributing to the research community what may help utilize the great potential of PSI. First is presented detailed electrochemical investigations into the role of dissolved O2 as a catalyst for methyl viologen (MV) to scavenge photoactivated electrons from PSI monolayers. These measurements, apart from demonstrating the ability of dissolved O2 in the electrolyte medium to act as a direct electron scavenger, also reveal that the dissolved O2 forms a complex intermediate species with MV which plays the essential role in mediating redox pathways for unidirectional electron transfer processes. Second, a novel 3D architecture is described to organize and stabilize PSI in the myriad of harsh conditions in which it needs to function. PSI is encapsulated in a highly stable nanoporous metal-organic framework, ZIF-8, denoted here as PSI@ZIF-8. The ZIF-8 framework provides a unique scaffold with a robust confining environment for PSI while protecting its precisely coordinated chlorophyll networks from denaturing agents. Pump-probe spectroscopy confirms the photoactivity of the PSI@ZIF-8 composites even after exposure to denaturing agents and organic solvents. This work provides greater fundamental understanding of confinement effects on pigment networks, while significantly broadening the potential working environments for PSIintegrated bio-hybrid materials. Finally, inspired by our successful encapsulation of PSI@ZIF-8, cations from this precursor are used to form charge transfer complexes with the extremely strong organic electron acceptor TCNQ. This PSI-Zn-H2mim-TCNQ charge transfer salt complex was dropcast on ITO as dense films. Through voltammetric cycling and the exchange of Zn2+ and H2mim+ cations, electrochemical annealing of the film increases electrical connectivity and electron conductivity that enabled PSI embedded charge transfer film to generate an order of magnitude greater photocurrents.

Ranchers, farmers, land managers, and resource management agencies have been plagued with various types of invasive weeds for decades, ultimately affecting millions of acres of viable wildlife habitat, grazing, and recreational opportunities. Invasive weeds often have nutritional contexts that negatively affect animals in some way, increasing the incentive to avoid consuming it by various animal species and domestic livestock. With the current amount of land infested with this weed, and the limited knowledge of the reason for avoidance, the underlying causes of livestock avoidance require in-depth and fundamental understanding. In my MS program, I sought to explore the relationship of plant and herbivore interactions in regards to grazing, animal diet selection through the use of supplementation, along with understanding the chemical compounds that cause avoidance of medusahead by domestic animals. This research will help us better understand the reasoning behind grazing avoidance and preference, and provide further understanding and management approaches using livestock as an alternative management tool in hopes of creating sustainable ecosystems with complementary benefits for soils, plants, animals, and land managers.

Abstract: The presence of 3-nitrotyrosine (nTyr) in proteins and peptides serves as a marker for the production of reactive nitrogen species. Nitrotyrosine occurs in cells in vivo at normal, basal levels, and in increasing amounts during states involving oxidant stress, such as inflammation. Tyrosine nitration can change the activity of many enzymes and is believed to occur in a site-specific manner. Tandem mass spectrometry provides the definitive method for identifying the presence and site of nitration, but the amounts of nTyr in vivo are often below detectable levels. To address this need we developed methods to specifically concentrate proteins and peptides containing nTyr to facilitate the identification of the site of modification. The first series of methods focused on specific, solution-phase labeling of dithionite-reduced nitrotyrosine with an amine-reactive biotin label (sulfo-NHS-S-S-biotin). After protein digestion, biotinylated peptides were affinity-captured with streptavidin-bound Sepharose and eluted by reducing the disulfide linker on the biotin tag. Although nitrated peptides from tetranitromethane-treated bovine serum albumin could be isolated with several variants of the method, all required very large amounts of sample to yield suitable data. A solid-phase capture approach was developed that used similar chemistry and also took advantage of the lower pKa of aminotyrosine to increase selectivity of binding. This yielded a 10-fold increase in sensitivity. The best implementation of the method, however still suffered from inadequate sensitivity and reproducibility. Of the two methods presented, however, it holds greater potential. Given the amount of non-specific reactivity of amines with the matrices that were used, it may he necessary to develop a more specific capture chemistry. The substrates also may have difficulty accessing reactive sites on the resin. Further optimization of binding conditions might yield a more robust and reproducible capture reaction. Given the methods developed, and with potential improvements to the reaction conditions, solid-phase capture appears to be a promising method to isolate nTyr-containing peptides and proteins.
