Conservation efforts on the Wild & Scenic North Fork American River stretch back over a century. Private landowners sought to protect wildflowers, wildlife and the watershed from development. At the same time, they sought to shape or maintain the landscape of the river in accordance with their own aesthetic preferences. Diverse groups sought to establish the river as Wild & Scenic in the 1970s. In recent decades, land trusts have purchased significant acreage in the North Fork in a push for broader conservation and public access. Why? More specifically — How have historic, economic, legal and aesthetic structures shaped the acceptance and growth of conservation in the North Fork? This project utilizes numerous methods in attempting to answer that question — archival fieldwork, interviews, representation of place through time, GIS, cartography, comparative case study analysis and muddy boots fieldwork. The project operates under an overarching theoretical framework built on a posthumanist political ecology approach that decenters human concerns as primary, focuses on political/power relations and their effect on biophysical reality and argues for a conception of the world that accounts for the equitable consideration of more than human entities. The approach both broadens the acceptance of who matters and deepens potential understanding of places and their associated landscapes. It also calls for greater humility in our approach to the entities with whom we share the Pluriverse and increased attention to the rights of the more than human world. This approach does more than help us understand the current state of the North Fork American River. It also allows us to see the importance of conservation efforts to the future health of all forms of life, including our own.

An Epidemiological Study of Campylobacter Populations Reveals a Selective Pressure by Roxarsone
Whole genome sequencing (WGS) is the most discriminatory of typing methods and can provide additional information including virulence, antigen targets for vaccine development, and antimicrobial resistance profiles. The first part of this study aimed to determine the application of WGS as a genotyping method for Campylobacter by comparing WGS to two commonly used genotyping methods, Pulsed-field gel electrophoresis (PFGE) and flaA typing. Five Campylobacter strains isolated from conventional and organic poultry, and five additional isolates with published genomes were compared using the three methods. A total of 8 PFGE patterns and 8 flaA alleles were identified from the 10 strains. Not surprisingly, WGS was more discriminatory than the other two methods and identified distinct differences among all ten isolates including genome size, G-C% content, and distribution of genes. Additionally, WGS revealed a number of virulence factors, including antibiotic and heavy metal resistance. Interestingly, genes coding for resistance to antibiotics were present in both conventional and organic isolates, but only conventional isolates possessed resistance to arsenic. The second part of this study aimed to understand the differences in resistance in organic and conventional isolates and determine if roxarsone, an organoarsenical coccidiostat used in poultry production, selected for arsenic resistance in Campylobacter. Eighty-two isolates of Campylobacter were collected from 246 retail organic and conventional chickens from 2009 to 2010 prior to the roxarsone withdrawal. In 2013 and 2014, an additional 65 isolates were collected from 120 retail organic and conventional chickens, after the roxarsone withdrawal. PCR determined the presence of arsenic resistance (ars) genes arsC, acr3, arsB, arsP, and arsR and agar dilution MICs were conducted to determine levels of resistance. Presence of arsP, arsC, and acr3 were significantly correlated to isolates from conventional chickens pre-roxarsone withdrawal (Pre-RW) and these isolates had higher resistance to all arsenic compounds tested, compared to those from organic. Post-RW isolates retained arsenic resistance genes, but an overall reduction in arsenate and roxarsone MICs was observed. It appears that despite the removal of the selective pressure of roxarsone, Campylobacter retains genes that confer arsenic resistance.

The headwaters basin of the North Fork of the American River, a roughly 20,000 acre valley high in California''s Sierra Nevada, reveals a case study of maintaining access to public lands in an area where a mixture of public and private ownership overlays the landscape. For those who seek to visit the area, navigating the web of public and private lands, public easements and navigable waterways presents a significant problem, as a rather contentious situation exists between private land ownership groups and the general public. There are ongoing efforts by owners in the area to prevent public use of trails whose existence pre-dates current ownership and probably pre-dates Anglo exploration of the area by a wide margin. This problem is exacerbated by the fact that these trails are at the bottom of a steep and rugged river canyon and, as such, no practical alternative exists for accessing the public lands both above and below the contested area. Through visitation of the area, interviews of relevant parties, and archival research, a clearer picture of the situation will be attained. At stake are not just recreational opportunities, but the right of the public to maintain historical use of travel corridors and access to publicly held land in spite of exclusionary practices by private landholders.
