DIEL REGULATION OF METABOLIC FUNCTIONS OF A WESTERN LAKE ERIE MICROCYSTIS BLOOM INFORMED BY METATRANSCRIPTOMIC ANALYSIS
Despite the global prevalence and apparent increased frequency of freshwater cyanobacterial harmful algal blooms (cHABs), metabolic functions that promote cHAB formation and maintenance remain poorly understood. An opportunity to elucidate cellular responses over diel cycles was presented following the 2014 Toledo (OH, USA) water crisis as part of a 48-h Lagrangian survey following toxin-producing bloom biomass in western Lake Erie. Taxonomic analysis of samples collected from 1 m depth showed the bloom to be dominated by three major phyla: Cyanobacteria (~30%), Proteobacteria (~30%), and Bacteroidetes (~20%). Differential expression analysis of Microcystis reads revealed strong diel patterns of expression between day (10:00 h, 16:00 h) and night (22:00 h, 04:00 h) samples. Day transcripts were abundant in functions related to photosynthesis (e.g. psbA, cpcB), nutrient and energy metabolism (glnA, nirA), cell division (ftsH), heat shock response (dnaK, clpB), and nutrient and ion transporters (bicA, acrB). Night transcripts included microcystin synthesis, amino acid synthesis (carB), and pre-dawn expression of photosynthetic antennae (chlI, cbiE). Hierarchical clustering and PCA showed a uniform grouping of expressed genes at night, whereas day transcripts were separated over the 48-h duration. Lack of uniform clustering of day transcripts suggests that the functional partitioning of gene expression during a Microcystis bloom is not limited to circadian regulation, but also to physico-chemical changes within the environment.

Pathogen-specific T cells are an integral component of the adaptive immune response and essential for efficient protection against infectious diseases. Work in our laboratory has shown that the long-term preservation of antiviral CD8 memory T cells (CD8 TM) constitutes a protracted and pervasive remodeling process that culminates in their enhanced secondary reactivity and protective capacity. Integrated molecular, phenotypic and functional analyses identified the extracellular adenosine generation/signaling pathway as a possible key element for the regulation of secondary antiviral CD8 TM responses. We were able to show the preferential expression of CD73, an ecto-5''-nucleotidase that generates adenosine from AMP, on antiviral CD8 TM cells as compared to primary effector CD8 T (TE) cells. This led us to show that CD73 expression on CD8 TM cells precedes the acquisition of a central memory (TCM) phenotype, described as a subset of CD8 TM cells with increased protective and proliferative capacity in a recall response. We went on to show that CD73 expression by antiviral CD8 TM cells directly corresponds to enhanced recall responses and represents a novel marker for an intermediate memory subset along the TEM and T¬CM differentiation pathway. This prompted our complementary investigations of the antiviral T cell response in mice deficient for CD73. CD73-deficient mice exhibited normal primary antiviral CD8 T cell responses, virus control and memory generation yet demonstrated impaired secondary antiviral CD8 T cell immunity as evidenced by significantly reduced expansions of secondary CD8 TE cell populations upon rechallenge. Due to compromised survival of CD73-deficient secondary CD8 TE cells, these defects could be rescued in retroviral reconstitution experiments that specifically corrected enzymatic but not other CD73 functions. Thus, CD8 TM cells unexpectedly engage the extracellular adenosine generation/signaling pathway for optimization of secondary responses through harnessing of T cell-intrinsic CD73 activity. Our results define a novel, "pro-inflammatory" role for adenosine in the regulation of pathogen-specific T cell immunity, and identifies a novel pathway for prophylactic or therapeutic interventions aimed at bolstering immune protection against infectious disease.

In the last two decades, interest in narrative conceptions of identity has grown exponentially, though there is little agreement about what a "life-narrative" might be. In connecting Kierkegaard with virtue ethics, several scholars have recently argued that narrative models of selves and MacIntyre''s concept of the unity of a life help make sense of Kierkegaard''s existential stages and, in particular, explain the transition from "aesthetic" to "ethical" modes of life. But others have recently raised difficult questions both for these readings of Kierkegaard and for narrative accounts of identity that draw on the work of MacIntyre in general. While some of these objections concern a strong kind of unity or "wholeheartedness" among an agent''s long-term goals or cares, the fundamental objection raised by critics is that personal identity cannot be a narrative, since stories are artifacts made by persons. In this book, Davenport defends the narrative approach to practical identity and autonomy in general, and to Kierkegaard''s stages in particular.

The concept of "projective motivation" is the central innovation in Davenport''s existential account of the everyday notion of striving will. Beginning with the contrast between "eastern" and "western" attitudes toward assertive willing, Davenport traces the lineage of the idea of projective motivation from NeoPlatonic and Christian conceptions of divine motivation to Scotus, Kant, Marx, Arendt, and Levinas. Rich with historical detail, this book includes an extended examination of Platonic and Aristotelian eudaimonist theories of human motivation. Drawing on contemporary critiques of egoism, Davenport argues that happiness is primarily a byproduct of activities and pursuits aimed at other agent-transcending goods for their own sake. In particular, the motives in virtues and in the practices as defined by Alasdair MacIntyre are projective rather than eudaimonist. This theory is supported by analyses of radical evil, accounts of intrinsic motivation in existential psychology, and contemporary theories of identity-forming commitment in analytic moral psychology. Following Viktor Frankl, Joseph Raz, and others, Davenport argues that Harry Frankfurt''s conception of caring requires objective values worth caring about, which serve as rational grounds for projecting new final ends. The argument concludes with a taxonomy of values or goods, devotion to which can make life meaningful for us.
