The Effects Of Niraparib, A PARP-1/-2 Inhibitor, In Sensitizing Human Head And Neck Cancer Cells To Proton And Photon Radiation
Purpose: Radiotherapy (photon [XRT], proton [PRT]) in combination with targeted therapy is a promising less toxic strategy for patients with head and neck cancer (HNSCC). Niraparib is an orally bioavailable poly-(ADP-ribose) polymerase (PARP)-1 and PARP-2 inhibitor, which targeting DNA damage repair (DDR). Whether niraparib can influence the response of HNSCC cells to XRT or PRT, and the impact of niraparib on the relative biological effectiveness (RBE) of PRT versus XRT are unknown. We investigated these unknowns in human HNSCC cell lines of differing human papillomavirus (HPV) status. Methods: Cell lines HN5, SqCC/Y1 (HPV-), and UPCI-SCC-154, UPCI-SCC-152 (HPV+) were used. Cellular colony forming (assessed by plating efficiency), radiosensitivity (assessed by clonogenic cell survival assay), and radiation-induced DNA double-strand breaks (DSBs, assessed by 53BP1 foci) were determined. Niraparib (1 u00b5M) was given 1 h before irradiation. Results: Niraparib reduced colony formation in HN5 (85.7% relative to untreated) and UPCI-SCC-154 (69.3% relative to untreated) cells. Niraparib enhanced the radiosensitivity of all 4 cell lines at 0.1 cell survival fraction; and improved RBEs with versus without Niraparib were also observed (Figure 1). Niraparib prolonged the presence of DSBs at 24h after XRT or PRT in these cells (Figure 2). Conclusion: Niraparib enhanced radiosensitivity of 4 HNSCC cell lines to both PRT and XRT, with improved RBEs by a possible mechanism of Niraparib inhibited DDR. These data suggest that Niraparib may have potential to improve the efficacy of both PRT and XRT in patients with HNSCC. Further in vivo and mechanistic studies are warranted.

Does Financial Misconduct Affect the Future Compensation of Alumni Managers
We explore how an organization''s financial misconduct may affect pay for former employees not implicated in wrongdoing. Drawing on stigma theory we hypothesize that although such alumni did not participate in the financial misconduct and they had left the organization years before the misconduct, they experience a compensation penalty. Our results support this prediction. The stigma effect increases in relation to the job function proximity to the misconduct, recency of the misconduct, and an employee''s seniority. Collectively, our results suggest that the stigma of financial misconduct could reach alumni employees and need not be confined to executives and directors that oversaw the organization during the misconduct.

Compressible Flow Structure of Droplets Under Supersonic Conditions
The compressible flow structure in front of deforming droplets in a continuously accelerating supersonic flow was examined in a draw-down supersonic wind tunnel which allowed relative velocities as high as Mach 1.8. 100-micron diameter droplets were generated upstream of the tunnel entrance using a Droplet-On-Demand generator. Direct backlit and schlieren imaging were performed in order to verify the experimental configuration and image the shock structure, respectively. The test liquid employed was 2-propanol due to its non-volatile nature and ease of injection. Theoretical predictions provide a first order approximation for several bow shock parameters. The schlieren images were processed to highlight the droplet and bow shock structure. Measurements of blurred shock thickness and Mach angle were found to be generally consistent with theoretical predictions. The time scales of shock formation and shock reaction to droplet deformation were estimated to be much less than the time scale associated with droplet breakup.

Text Mining of Online Book Reviews for Non-trivial Clustering of Books and Users
The classification of consumable media by mining relevant text for their identifying features is a subjective process. Previous attempts to perform this type of feature mining have generally been limited in scope due having limited access to user data. Many of these studies used human domain knowledge to evaluate the accuracy of features extracted using these methods. In this thesis, we mine book review text to identify nontrivial features of a set of similar books. We make comparisons between books by looking for books that share characteristics, ultimately performing clustering on the books in our data set. We use the same mining process to identify a corresponding set of characteristics in users. Finally, we evaluate the quality of our methods by examining the correlation between our similarity metric, and user ratings.

Distinct N-terminal Domains Regulate PSD-MAGUK Conformation and Selection of Binding Partners
PSD-95-like membrane associated guanylate kinsases (PSD-MAGUKs) are multidomain scaffolding proteins involved in AMPA and NMDA receptor trafficking and clustering at synapses. As proteins with multiple protein-protein interaction domains, they have a capacity to interact with many integral membrane receptors, cytoskeletal components, and signaling molecules although how these interactions are regulated is unclear. Multiple conformational states for some PSD-MAGUKs have been described and could act to regulate the availability of their individual domains. However, an in vivo demonstration of these conformations, especially in neurons, and an understanding of what defines their conformational states is lacking. In the work presented here, conformations of PSD-MAGUKs and how they are regulated were inverstigated using a FRET-based approach where the N- and C-termini of SAP97beta, PSD-95alpha and SAP102 were tagged with a mCherry and Venus FRET pair. By measuring changes in intramolecular FRET, we demonstrate in HEK293 cells and in neurons that PSD-MAGUK proteins alter between "compact" and "extended" conformations. Further, we reveal that interactions between PSD-MAGUKs and different NMDAR and AMPAR subunits are dependent on PSD-MAGUK conformation. These studies also find that transitions between conformational states are regulated by changes at different sites at the PSD-MAGUK N-termini, the L27 protein interaction domain (SAP97beta), the palmitoylation site (PSD-95alpha), and the zinc-binding domain (SAP102). Parallel electron microscopy measurements at the PSD confirm and extend the conclusions about PSD-95alpha conformation based on FRET measurements. Our results reveal the dynamic nature of the structure of PSD-MAGUKs, identify a novel regulatory role for their N-termini, and provide mechanistic insight into how PSD-MAGUK family proteins determine their selection of different binding partners.
