Supply chains in the pharmaceutical industry are growing increasingly more complex and expanding their geographic reach both in manufacturing production and to the end consumer, the patient. Physical development, manufacturing and distribution of these drugs, both of biologics and small molecules, is extremely technical in science and processes. Additionally, the industry is highly regulated with nuanced requirements that vary by country of origin and consumption, adding complexity to the drug development process. For these reasons, companies are pushing for longer range planning and forecasting of their drug pipelines, beginning the process earlier for drugs that are in pre-clinical phases of production in order to adequately plan for capacity in manufacturing and distribution. Working with data on a number of small molecules across different lines of treatment in the drug development pipeline, a discrete event simulation model was developed to simulate production quantity outputs given varying levels of stochastic parameters such as drug dosage, treatment duration, patient population, patient compliance, and competitive market share. Results from the simulations were used to assess manufacturing capacity risk given capacity and resource capabilities. The outputs of the model built in this thesis can be used to better inform capacity planning decisions for these early stage molecules.

The Role of Smaug in Post-transcriptional Regulation
Smaug is a sequence-specific RNA-binding protein (RBP) and a multifunctional post-transcriptional regulator. In this thesis, I have used genome level analysis to gain a panoramic view of regulation by Smaug during early Drosophila embryogenesis. I show that Smaug plays a direct and major role in the translation and stability of a large set of maternal mRNAs. Smaug''s target transcripts function in a diverse array of processes including metabolism, lipid droplet function, protein folding and protein stability, suggesting previously uncharacterized functions for Smaug. I also performed detailed analysis of Smaug''s regulation in the embryo''s germ plasm and found that Smaug sits at the top of a posttranscriptional regulatory cascade that controls primordial germ-cell number by attenuating synthesis of the embryonic germ plasm. In the germ plasm, Smaug binds to arrest mRNA to repress the translation of Bruno (BRU) protein. BRU potentiates the production of Oskar (OSK) protein and germ plasm, Thus, repression of Arrest/Bruno (Bru) by Smaug is required for the establishment of the correct number of germ cells during early embryogenesis.

Detecting Sources of Heat Loss in Residential Buildings from Infrared Imaging
Infrared image analysis was conducted to determine the most common sources of heat loss during the winter in residential buildings. 135 houses in the greater Boston and Cambridge area were photographed, stitched, and tallied to characterize nine major causes of heat loss: window frames, window surfaces, window cracks, basements, door cracks, corners, chimneys, roof ridges, and soffits. The nine causes of heat loss were mapped to the three modes of heat transfer: conduction, convection, and radiation. It was found that heat losses through window surfaces, window cracks, chimneys, and soffits dominated as common sources of energy leakage, each represented in more than 70% of the houses analyzed. Opportunities for future work include more thorough examination of losses through ducts and walls, as well as developing methods for improvements.

The Use of a Computer-Based Writing Program
The invention of computer-based writing program has revolutionized the way of teaching second language writing. Embedded with artificial intelligence scoring engine, it can provide students with both immediate score and diagnostic feedback on their essays. In addition, some of such programs offer convenient writing and editing tools to facilitate students'' writing process, such as MyAccess. However, the effectiveness of using such programs to improve students'' writing skills in the L2 is not conclusive yet. To gain a deeper understanding of how such a computer-based writing program works in EFL writing classes, this study explored factors that may lead to facilitation or frustration when the program MyAccess is in use. The participants were three EFL writing classes consisting of 68 third-year English majors in a Taiwan''s university. Data included students'' responses to a questionnaire, writing samples, feedback given by MyAccess, and focus-group interviews with the students. The findings show that only 55% of the students felt this program either moderately or slightly helpful to them in improving their writing skills and that the students were much less satisfied with the grading function than the writing/editing functions. Their dissatisfaction was largely because this program failed to give specific feedback in the content and rhetorical aspects of their writings. Moreover, the study also found that the different ways the three writing classes used this program and the instructors'' familiarity level with the program coupled with their technological skills greatly affected the students'' attitudes toward this program. The study concludes that pedagogical approaches and contextual factors are more crucial than the functions of the program in determining its effectiveness. (Contains 5 tables and 1 figure.).

From a social-interactionist''s view of language learning, "reflective conversation" via CMC, particularly through the asynchronous mode, has been advocated (Lamy & Goodfellow, 1999). It emphasizes learning through reflection on experience and through the exchange of thoughts with others. Guided by this pedagogical model, asynchronous discussion was implemented in an EFL content-based course taught twice in 2003 and 2004 that integrated learners'' language learning experience with language learning theories. The study investigated the effectiveness of learning both content and language in this experience-based language learning course through asynchronous discussion and also examined the reflectiveness and interactivity of the participants'' online exchanges. The results showed that most of the participants held a highly positive view on this CMC learning activity. Moreover, their reflectiveness in asynchronous discussions was found to be affected more by peer interaction than by the instructor''s participation. Of particular interest is that the differences found among discussion groups revealed that the higher participation level the students had, the more positive view they held. This can be well supported by Wenger''s social theory of learning emphasizing the importance of active participation.
