Remarkably, all such co-implants suppressed splenocyte proliferation at 10 days post-implant, though this effect disappeared by 60 days. In contrast, the antibody response was equal to or greater than that for a skin graft alone at both timepoints. Th1/2 polarization did not explain these observations, since it did not correlate with suppression of splenocyte proliferation, but rather depended on mode of xenogeneic cell delivery. Microencapsulation has potential clinical application for islet transplantation. To better understand the ongoing destructive host response, I examined whether the adaptive immune system recognised encapsulated xenogeneic cells, and how immune responses interacted with implanted biomaterials. The influence of Th1/2 bias did not explain the observed suppression. Encapsulated xenogeneic cells elicited a consistent Th2 response, unencapsulated cells elicited a mixed response, and skin xenografts showed an initial Th2 response that transformed to a Th1 response by 60 days based on antibody isotypes and cytokines. These novel effects of biomaterials have wide relevance: most tissue-engineered constructs contain biomaterials and will elicit immune responses, so suppressions mediated by biomaterials merits further examination and should affect future design approaches. I then monitored host immune responses to skin xenografts accompanied by one of three co-implants: encapsulated xenogeneic cells; unprotected cells accompanied by the encapsulation biomaterials; and encapsulation-biomaterials without cells. Encapsulation did not prevent a strong host antibody response (day 10 and day 60). Splenocyte proliferation in vitro did not differ whether primed by implanted, encapsulated cells, or unencapsulated cells (60 days). Thus, capsule membranes did not prevent indirect recognition of shed antigens. However, splenocyte proliferation to these cell/biomaterial implants was delayed compared to skin xenografts: after 10 days implantation, proliferation was dramatically lower than to skin grafts, though the difference disappeared by 60 days. This transient T-cell suppression was unexpected, since encapsulated cell viability was already compromised by 10 days. Mice were implanted intraperitoneally with encapsulated and unencapsulated xenogeneic cells, and the responses were compared with xenografted skin (positive control). Splenocyte proliferation upon re-challenge in vitro, antibody titre in serum, and Th1/2 polarisation (by IL-4 and IFN-gamma in splenocyte challenge supernatants and IgGl and IgG2a antibody isotypes in serum) were measured.

The purpose of this review is to evaluate the clinical and cost-effectiveness of hyaluronic acid (HA) relative to other knee osteoarthritis (OA) interventions, and to review evidence-based guidelines for the use of intra-articular (IA) hyaluronic acid (HA) for knee OA. There is a substantial body of evidence investigating viscosupplementation with hyaluronic acid (HA) in adults with knee osteoarthritis (OA), but the evidence to support its efficacy is conflicting. Three meta-analyses rated by investigators as the best available evidence suggested that HA was superior to intra-articular (IA) placebo, corticosteroids (CS), and non-steroidal anti-inflammatory drugs (NSAIDs), in terms of improving knee pain and function, without increasing adverse events. However, another study reported that when only the highest-quality randomized controlled trials (RCTs) were meta-analyzed, no clinically important differences of HA treatment over IA placebo were observed. There are significant limitations reported in the HA literature, including flawed study designs and reporting, a strong placebo effect from IA injections, potential conflicts of interest due to industry funding, and variation in dose, treatment course, and molecular composition of the various HA agents under study. It has been suggested that statistical significance of effects observed in some HA studies may not have clinical significance. Limited high-quality evidence suggests that two types of HA agents are cost-effective in the treatment of knee OA compared with other interventions including NSAIDS and other analgesics, as well as other forms of conservative care such as physiotherapy, weight loss, and ambulatory aids. The majority of guidelines did not find sufficient evidence to make a recommendation for or against the use of HA for knee OA, however two guidelines recommend against its use. Some guidelines recommend HA after failure of other treatments, or in older adults with a certain OA grade.

Phatic communion, or "small talk," has long been overlooked in literature on both pragmatics and language learning. This project examines L2 phatic competence by looking at how adult English language learners (ELLs) engage in phatic communion outside the classroom, and how they identify and interpret phatic interactions. Naturalistic data were collected from unstructured conversations in "conversation pubs." The data were analyzed from an interactional standpoint, with a focus on performativity and a view to what strategies participants employ to engage in phatic communion, and were supplemented with semi-structured interviews and the researcher's own background knowledge of the participants. Phatic communion came to be defined by the participants themselves, and is enacted in this study through a variety of speaker strategies and indexical cues. This project aims to provide valuable data regarding L2 phatic use and interpretation, with implications for researchers, language educators and professionals.

The Role of Transcription Factors in Regulating Adult Neurogenesis After Excitotoxic Brain Injury
While compensatory striatal neurogenesis is well documented in many injury models, cells of the correct lineage for endogenous repair are not always regenerated. To understand why, the molecular profile of subventricular zone (SVZ)-derived neural progenitor cells (NPCs), and their response to injury was investigated. Expression of the transcription factors Mash1, Dlx2, Pax6 and Olig2 in transitamplifying- precursor cells (TAPs) and neuroblasts was examined following quinolinic acid (QA) lesioning of the adult rat striatum. It was demonstrated that TAPs responded to lesioning in a heterogeneous manner, with expansion particularly of the Dlx2+ subpopulation. Dlx2+ neuroblasts also increased at early time points compared to controls. Previous work identified SVZ-derived bipolar and multipolar cells within the lesioned striatum. I characterised and quantified migration and differentiation of retrovirally-labelled SVZ-derived green fluorescent protein (GFP) cells within the lesioned striatum. In addition to neuroblasts, migratory Olig2+ bipolar cells were observed, representing oligodendrocyte progenitor cells (OPCs). Multipolar cells also expressed NG2, indicating OPCs were recruited into the lesioned striatum. Proneural genes can repress Olig2 lineage cells, and substantially increase neurogenesis when overexpressed in the adult brain. Proneural Dlx2-GFP and Pax6-GFP retroviruses were injected into the SVZ on the day of lesioning, or two days post lesion (dpl), when the greatest migratory response was observed in controls. Surprisingly, Pax6 overexpression decreased striatal neurogenesis from NPCs born on the day of lesioning, but increased oligodendroglial cells from NPCs born two dpl. Dlx2 overexpression had no effect on NPC migration from cells born at the time of lesioning. However at two dpl, a non-neurogenic time point in controls, Dlx2 increased numbers of striatal neuroblasts and the percentage of neurogenic cells in the lesioned striatum. These results indicated preferential recruitment of Dlx2+ NPCs, GABAergic lineage precursors and the correct cell type for QA-induced striatal regeneration. However, retrovirallyoverexpressed proneural genes were downregulated within the lesioned striatum, suggesting factors secreted from injured areas could alter cell fate. Quantitative-PCR analysis of signalling genes confirmed multiple SVZ alterations after injury, some with the potential to increase oligodendrocytelineage signalling. Understanding molecular signals regulating lineage after injury is essential before regenerative medicine can progress towards successful therapies.

A Cognitive Approach to Understanding One's Emotions
I use an analytical approach to understanding my cognitive and emotional responses. I study how my thoughts and memories can affect my frame of mind, sometimes taking me down a negative mental path. I look at my past to understand the responses behind my current motives and to better understand myself. This recollection of certain memories becomes complex ; I see my head being filled with a multitude of emotions. Constantly swirling in my mind. This can lead to a place of vulnerability, which I try to conceal from the people around me. Sometimes, the vulnerable thoughts lead to exposing my emotional state. My immediate response is to conceal my emotions from others ; but at times there is little else to do but to reveal them. I choose materials that echo a sense of fragility like that of a decayed leaf. That sense of delicacy is like that of a fragile thought which can take over your mind and consume you. I look at natural occurrences like tornadoes whose swirling movement is akin to the disruptions I experience in my patterns of thinking. I see influence in strata of rocks that represent the layers of thoughts from an event that has shaped me. Ultimately, I'm trying to better understand myself and the world around me.
