Differentitiation or decidualization of the human endometrial stroma is controlled by steroid hormones. Our aim was to study prolactin (PRL) gene expression, which occurs concomitant with decidualization, by DNase 1 hypersensitivity analysis and transient transfection analysis. Two endometrial stromal cell lines, B10T1 and M4, treated with 8-Bromo-cAMP (cyclic adenosine monophosphate) and MPA (medroxyprogesterone acetate), did not produce PRL. Using lymphoblast cell lines IM-9-P33 (positive-PRL) and IM-9-P6 (negative-PRL) for DNase I hypersensitivity, no sites were detected in 22.5 kb of decidual PRL 5$\sp\prime$ flanking DNA. Intron A-1 contained three sites in IM-9-P33. PRL promoter deletion constructs containing 5000 bp of 5$\sp\prime$ flanking DNA and deletions therein transiently transfected into IM-9-P33, IM-9-P6, non-treated and treated B10T1 and HeLa cell lines revealed basal activation. Transient transfections of the region into the same cell lines revealed basal activation in the sense orientation and 60-80-fold increase when placed in the antisense orientation.

Temperament in cattle is often defined as the reactivity to human handling or novel environments. Temperament differences have been shown among breed-type categories, within breed types, among crossbreds, and between sexes. Temperament tests are typically completed at weaning time on beef cattle, and rarely on fed Holstein steers for beef production. Balking behavior, or cease in forward motion, in the cattle working facility can pose welfare issues as the electric prod use to coerce movement is implemented. Three observational field projects were designed to evaluate balking behavior incidence in unknown breed-types at the processing plant, Holstein steers in the feedlot and processing plant, and Angus and Hereford-Angus crossbred steers over their lifetime. Issues related to the timing and nature of change in treatment research studies are important ones that have not been addressed adequately in the alcohol dependence treatment literature. The current study applied both Early Treatment Response (ETR) and Sudden Gains (SGs) methodologies to a sample of women who sought treatment for alcohol dependence. An ETR refers to a greater response in less time than would be expected (e.g., 65% decrease in depression in the first four weeks of a twenty week treatment protocol). SGs, on the other hand, are \"sudden, substantial symptom improvements\" that occur between two sessions and are hypothesized to be the result of what occurred in the first of those two sessions. SGs can happen at any time during the treatment trial (and can happen more than once) and are individualized (i.e., they do not necessarily occur for the whole sample nor, for those who do experience SGs, at the same time). The overall goal of the study was to explore the utility of these methodologies in an alcohol treatment population.
