Human papillomaviruses (HPVs) are small non-enveloped DNA viruses that are causally associated with multiple human cancers. As HPVs have strict epithelial tropism, primary human keratinocytes are generally accepted as physiologically relevant host cells for HPV infection. However, these cells exhibit very low permissivity to HPV infection in comparison to immortalized keratinocytes. We hypothesized that HFKs have more robust host defense mechanisms to restrict HPV infection compared to immortalized keratinocytes. Here, these studies show that the basal level of autophagy in primary human foreskin keratinocytes (HFKs) is higher than in immortalized keratinocytes, and that HPV virions significantly induce autophagy in HFKs. Autophagy, a cell-intrinsic pathway that engulfs and degrades bulk cytoplasm, has important roles in cell homeostasis as well as host defense against invading pathogens. This study shows that HPV infectivity is dramatically enhanced by knockdown of essential autophagy genes as well as biochemical inhibition of autophagy in HFKs. Furthermore, inhibition of autophagy delays degradation of HPV capsid proteins during virus trafficking, indicating that host autophagy induced by HPV virions inhibits infection of primary keratinocytes through rapid degradation of viral capsid proteins. Further analysis to identify virus and host factors mediating autophagy induction by HPV revealed that HPV virus-like particles comprised of only the major capsid protein, L1, are sufficient to induce autophagy. Induction of autophagy by HPV capsids is mediated by CD46-GOPC signaling, a pathway which has recently been reported to induce autophagy in response to measles virus and group A streptococcus. This work identifies host autophagy as a restriction factor against HPV infection. Induced by HPV capsids and CD46-GOPC signaling during entry, autophagy may inhibit HPV infection by facilitating capsid protein degradation of incoming virions. Finally, this study also shows that the anti-viral inferferon-inducible transmembrane proteins (IFITMs) do not inhibit HPV infection in HFKs. As IFITMs inhibit cytosolic entry of several viruses requiring low pH for infectious entry, these results indicate that the entry mechanism of pH-dependent HPV virions bypasses IFITM restriction.
