Historian Ines Prodöhl describes the soybean’s journey centered on three hubs: Northeast China, as the crop’s main growing area up to the Second World War; Germany, to where most of the beans in the interwar period were shipped; and the United States, which became the leading cultivator of soy worldwide during the 1940s. Innovative Small-molecule Therapies for Psoriasis
Psoriasis is a common, chronic inflammatory skin disease with a strong genetic predisposition. Formerly considered a purely skin disease, it is now recognised as a chronic systemic condition that may affect the nails and joints and can increase the risk of coexisting chronic conditions, such as obesity and cardiovascular disease. The majority of patients experience mild to moderate severity of psoriasis, for which topical treatments are the first-line therapy. For more severe forms, ultraviolet therapy or systemic treatments, such as methotrexate and monoclonal antibodies, are available and can be highly effective. Despite these options, some patients experience insufficient results or adverse effects, and psoriasis is well established to significantly reduce health-related quality of life. In 2014, the World Health Assembly Resolution WHA 67.9 emphasised the need for new, widely accessible treatments for psoriasis. The aim of this doctoral thesis was to evaluate novel small-molecule treatment options for psoriasis within the field of redox medicine. In Study I, we investigated small-molecule MutT Homolog 1 (MTH1) inhibitors in preclinical models. The enzyme MTH1 is crucial for preventing oxidative damage to guanine, a nucleotide and building block of DNA. Cancer cells exhibit increased oxidative DNA damage and over-express MTH1 to protect their survival. Inhibition of MTH1, a strategy to disrupt this protective mechanism, is currently under clinical investigation by Prof. Helleday''s group at Karolinska Institutet (KI). Psoriasis shares pathological features with cancer, such as hyperproliferation, a pro-inflammatory environment, and angiogenesis, although malignant transformation generally does not occur in psoriatic cells. Psoriatic tissue also exhibits disrupted redox homeostasis, and we demonstrated in Study I that the enzyme MTH1 is upregulated in human psoriatic skin. MTH1 inhibition with subcutaneous injections reduced histological characteristics and inflammation in an in vivo mouse model of psoriasis. Our results of genetic analyses further support MTH1 inhibitors as a potential future treatment option for psoriasis. In Study II, we investigated the functional role of the gene nitric oxide synthase 2 (NOS2), which has been identified as a susceptibility gene for psoriasis. It has also been suggested as a disease classifier, distinguishing it from different types of eczema, another common skin disease. The small molecule nitric oxide (NO) is produced by NOS2, and other isoforms of NOS. NO has been shown to exhibit pleiotropic effects depending on its concentration in cells, which can lead to opposite effects on various biological functions, such as promoting or inhibiting proliferation and inflammation. We demonstrated that NOS2 contributes significantly to the psoriatic phenotype in a mouse model of psoriasis. Additionally, we observed beneficial effects of an NO-donor cream in the imiquimod (IMQ) mouse model, with decreased keratinocyte proliferation and reduced inflammatory infiltrate in the dermis. The results of Study II highlight the therapeutical potential of an NO-donor cream as a novel topical treatment option for the majority of patients with mild-to-moderate psoriasis. In Study III, we evaluated a topical formulation of an MTH1 inhibitor (MTH1i TH1579) and demonstrated a diminished psoriatic phenotype in an in vivo mouse model of psoriasis. We showed that the number of IL-17-producing T-cells, crucial immune cells involved in driving the inflammation, was significantly reduced in murine skin following short-term treatment. Additionally, we observed a near abolition of IL-17 gene expression, an effect that may play a pivotal role in the pathophysiology of psoriasis. Furthermore, we explored an MTH1-S-phase kinase-associated protein 2 (SKP2)-Enhancer of zeste homolog (EZH2) pathway potentially involved in the antiproliferative effects of topical MTH1 inhibition through its impact on the cell cycle. In conclusion, this thesis provides the foundation for the development of novel treatment options for psoriasis with the small-molecule substances MTH1i TH1579 and NO, the latter incorporated into an NO-donor cream, showing beneficial effects in a well-established mouse model of psoriasis. Forskning under de senaste decennierna har visat att NO har olika funktioner beroende på dess koncentration. I studie II undersöktes därför betydelsen av NOS2 för psoriasis i cell- och djurmodeller. I mus-försök analyserades effekten av ökad NO genom applicering av en NO-frisättande kräm. En förbättring av den psoriatriska inflammationen observerades, vilket indikerar att behandling med en NO-frisättande kräm kan vara en möjlig framtida behandlingsstrategi mot psoriasis. Sammanfattningsvis banar de tre studierna i avhandlingen väg för innovativa behandlingsalternativ för psoriasis inom redoxmedicin, med särskilt fokus på topikala behandlingar som kan nå en större patientgrupp, där nuvarande behandlingsalternativ är begränsade.

Aim: The European Reference Networks (ERNs) provide clinicians and patients the opportunity to collaborate at EU level to improve diagnosis, care and treatment for people living with rare and complex conditions. However, building a partnership culture to systematically involve patients in ERN activities and decision-making structures is challenging, partly because the role of patient representatives and the value of this collaboration are not always understood. The objective of this project was to develop an evaluation framework to assess the impact of patient engagement in the ERNs and to provide evidence on the value of patient-clinician partnership. Methods: The evaluation was developed by EURORDIS and patient representatives involved in the ERNs (ePAG advocates) through a participatory and iterative process. The work was organised in three different phases: (1) clarify roles and identify common goals for ePAG advocates’ engagement in the ERNs; (2) define a set of measures; and (3) test the measures in three different ePAGs (European Patient Advocacy Groups). Results: The project allowed developing a common understanding among ePAG advocates of their role and goals in the ERNs and defining a patient-driven evaluation framework to assess their level of engagement in the ERNs’ activities and how effectively they were working to fulfil their role. Conclusion: Engaging with ERN clinicians to refine the framework would probably render it more relevant to the reality and priorities of the specific ERNs and more valuable as a tool to build a strong partnership culture. Such an evaluation framework could be integrated into the ERNs’ quality improvement system to ensure that the networks’ activities are driven by and remain responsive to patients’ needs.

People say, that it is nowadays essential for being successful in business to be a teamworker, a teamplayer. But what exactly is a team and what makes teamwork special? Several books and the internet helped me to gather relevant information in order to answer this question. Worldwide markets have changed enormously in the last two decades. Globalisation and liberalisation on the one hand and saturated markets on the other hand have led to high quality standards and customer orientation. In addition to that product life cycles have shortened, so that companies are under pressure to develop more new products and product variations in less time to reasonable costs. The need for more flexibilty and the necessity to adapt quickly to changing market conditions involved a reduction of layers in the management. One of the results is more responsibility for the employees. Not only that they become more and more involved in the decision making process, they also have to meet higher expectations as the tasks become increasingly more complex. It seems obvious that at a certain point one person alone can not handle difficult tasks any longer, especially if high financial and economic risks are associated. Therefore a rising number of companies forms teams. This essay focuses on aspects a team leader has to consider if he or she gets the task to start a team from scratch. While the first chapter has given a short introduction, the second chapter defines teamwork and the steps on the way from a mere group towards a team. Then the third chapter examines conditions under which teams can prosper and perform outstandingly. After that the fifth chapter gives a short summary and allows an outlook. The last three chapters contain a list of vocabulary, references and a test, where you can find out what kind of teamplayer you are.

This book introduces in a constructive manner a general framework for regression and fitting methods for many applications and tasks involving data on manifolds. The methodology has important and varied applications in machine learning, medicine, robotics, biology, computer vision, human biometrics, nanomanufacturing, signal processing, and image analysis, etc. The first chapter gives motivation examples, a wide range of applications, raised challenges, raised challenges, and some concerns. In order to create the optimally standardized bodies that cosmonauts would require, top secret military laboratories were set up in 1970s East Germany. The New Man – the modern colonist of space – was intensively trained for the purpose of surviving years of weightlessness in outer space. Experiments were carried out in prisons, hospitals and army barracks with the aim of creating the perfect body: self-sufficient and able to endure extreme conditions for as long as possible. In order to exert dominance over space, it was first necessary to exert total control over those who were being trained to conquer it. Ines Geipel unravels this largely unknown and extraordinary history by delving into East German military records and talking to those who bear the scars of this state-inflicted trauma.
