The computers visual representation of sound has revolutionized the creation of music through the interface of the Digital Audio Workstation software (DAW). With the rise of DAW-based composition in popular music styles, many artists sole experience of musical creation is through the computer screen. I assert that the particular sonic visualizations of the DAW propagate certain assumptions about music, influencing aesthetics and adding new visually-based parameters to the creative process. I believe many of these new parameters are greatly indebted to the visual structures, interactional dictates and standardizations (such as the office metaphor depicted by operating systems such as Apples OS and Microsofts Windows) of the Graphical User Interface (GUI). Whether manipulating text, video or audio, a users interaction with the GUI is usually structured in the same mannerclicking on windows, icons and menus with a mouse-driven cursor. Focussing on the dialogs from the Reddit communities of Making hip-hop and EDM production, DAW user manuals, as well as interface design guidebooks, this dissertation will address the ways these visualizations and methods of working affect the workflow, composition style and musical conceptions of DAW-based producers.

The gap between the achievement scores of Latino and Black students compared to the scores of White and Asian students has received considerable attention from educators, with little progress made toward closing the gap. This study used growth mindset research to address four destructive learning threats that may contribute to the achievement gap for students in high-minority, low-income schools: negative school culture norms, the Pygmalion effect, stereotype threat, and fixed mindsets. Since the publishing of growth mindset research, leaders and teachers have looked for guidance on harnessing its ability to close achievement gaps in schools. Interventions for removing these threats to learning show promise. For this qualitative participatory action research study, I designed the growth and fixed mindset norms framework for implementation in school cultures. The framework was created to give school leaders a tool for building school culture norms that counteract the learning threats. Seven school leaders participated in three reflection, learning, and planning sessions, which gave them the tools to implement the norms. School leaders were given autonomy to choose the best path toward implementing the framework at their schools. Leaders conducted pre- and post-session observations of school cultures using the framework with the intention of measuring success levels for norm change. I analyzed the leaders'' implementation actions, which revealed drivers and barriers to implementation of the norms. One leader achieved high norm change, while three leaders achieved moderate norm change. Their actions illuminated a set of strategies for implementation. The successful leaders provided professional development and coaching on the norms. However, the most successful leaders provided teachers with targeted coaching on the norms and offered intervention coaching for struggling teachers. The four unsuccessful leaders did not provide their teachers with these same competency-building activities. Instead, they claimed they were stalled in their desire to implement the norms by shared-decision making models. As a result, their schools showed no norm change or their school cultures became more fixed over time. The findings in this study show that norm change is possible with intentional and coordinated action. Based on the experience of the participating leaders, I provide recommendations for leaders seeking to build schoolwide cultures with growth mindset norms.

The objective of this work was to develop a biophysical model that would elucidate the role of the brain neurofibrillary tangles (NFTs) play in the development of Alzheimer''s disease (AD). AD is a devastating neurodegenerative disease, and this study [the study of Ian] is not only of substantial societal impact, but it will also contribute to the multidisciplinary efforts aimed at early detection, understanding factors behind disease progression and possibly personalized treatment of the disease. AD affects approximately 6.2 million Americans and is expected to grow exponentially by 2050 as the population ages. Those with AD required 15.3 billion hours of care, most of which is unpaid care from family members, and costs, on average, $321,780 per person.Current hypotheses on the mechanism of AD incorporate the combined role of two proteins: beta amyloid plaques and NFTs. They are hypothesized to work together in the amyloid cascade hypothesis to bring about the toxic effects of disease. The tau protein normally stabilizes microtubules in the central nervous system, but its phosphorylation causes destabilization and the formation of tangles. The direct cause of this phosphorylation is unknown, but amyloid plaques are believed to play a role. These tangles disrupt neuron function and synaptic communication. Unable to communicate, these cells die. The tangles propagate via synapticsynaptic connections, resulting in neuronal dysfunction, eventually cell death and thus regional atrophy which follows the pattern of NFTs. Cognitive decline increases as the disease worsens, generally thought to correlate with increasing spread of NFTs. Currently, AD is only able to be definitively diagnosed at autopsy, complicating the understanding of the disease and validation of therapeutic clinical trials. The advent of amyloid- and tau-specific PET tracers in conjunction with other imaging biomarkers has led to a hypothetical model of the progression of AD. However, the mechanism of propagation of disease is still not well defined.Quantitative and qualitative descriptions of the accumulation and advancement of NFTs throughout the cerebrum from both in vitro pathologic staining and in vivo flortaucipir NFT PET imaging characterizes the propagation of NFTs by increasing local intensity and increasing spatial extent. Here we describe a local accumulation connected spread (LACS) model of NFT propagation which naturally encompasses the intensity-extent phenomenon through a reaction-diffusion equation wherein the molecular mechanisms leading to tau accumulation in neurons represents the reaction and the expansion of tau with increasing disease stage along connected white matter trajectories is considered as a diffusion process.We explored three objectives to better understand the utility of the LACS model in predicting future NFT burden as represented by flortaucipir PET. First, we examined confounding factors inherent to flortaucipir PET and their effects on flortaucipir quantitation. Second, we generated a whole brain connectome to model the diffusion process and related harmonics of the connectome to flortaucipir uptake to understand concordance between connectivity and NFT burden. Third, we estimated parameters for LACS using Bayesian inversion to incorporate the uncertainties inherent to flortaucipir quantitation and the LACS model itself and predicted future NFT burden in a clinical trial population. The application of LACS modeling may provide insight into the accumulation and propagation of NFTs as represented by flortaucipir (FTP) PET throughout the cerebrum as well as the development of novel, tailored endpoints for AD therapeutic trials.
